Abstract
Research aimed at understanding DNA repair mechanisms in
both damaged and normal cells remains highly relevant. The
aim of this study was to investigate how activation of sirtuin 1
affects the distribution of DNA single-strand breaks in mouse
oocyte follicular cells. The alkaline comet assay was used,
in which, during electrophoresis of lysed cells embedded in
agarose gel, loops and fragments of damaged DNA migrate
toward the anode, producing a characteristic comet-like ap-
pearance. The length of the comet tail positively correlates
with the extent of DNA damage (nuclei were classified into
damage classes 0-4). Under conditions of oxidative stress, the
proportions of class 3 and class 4 nuclei increased by 2.6-fold
and 7.8-fold, respectively, whereas the proportions of class 0/1
and class 2 nuclei decreased by 18.6-fold and 2.3-fold, respec-
tively, compared with control cells. These findings indicate a
redistribution of DNA single-strand breaks in follicular cells
under oxidative stress. Resveratrol, a sirtuin 1 activator, was
found to promote DNA repair, as evidenced by an increase in
the proportion of cells with class 0/1 nuclei and a decrease
in the proportion of cells with class 3 and class 4 nuclei. The
obtained results provide evidence that sirtuin 1 activation
represents a promising strategy for reducing DNA damage in
mouse oocyte follicular cells.
