ROLE OF SIRTUINS IN CHANGE ON THE FUNCTIONAL STATUS OF OOCYTES AND CUMULUS CELLS OF FEMALE MICE UNDER CONDITIONS OF THE SYSTEMIC IMMUNE INJURY

Abstract

The effect of sirtuins activator (resveratrol) on the functional state of oocytes (meiotic maturation) and cumulus cells (apoptosis and necrosis) was assesed using the model of systemic autoimmune damage created by immunizing the CBA line mice with kidney antigen suspension. The conducted researches revealed significant ovarial damages of mice under the conditions of experimental model. A significant decreasement of number of mature follicles isolated from the ovaries was observed and an increasement (compared with control) of the number of oocytes with atypical morphology (unevenly granulated cytoplasm, signs of fragmentation of the cytoplasm) was noted. It was shown that, under experimental conditions during cultivation, the percentage of cells that dissolved germinal vesicle and capable to form first polar body significantly decreased. We have shown that inhibition of ovarian function was accompanied by a significant changes in viability and loss of cumulus cells. After the cultivation of oocytes in a medium which contained activator of sirtuins – resveratrol (20 μM), a significant increasement of the percentage of oocytes that dissolved germinal vesicle and formed the first polar body was detected, as well as the increasement of number of viable cells and the decreasement of the number of cells with morphological signs of apoptosis and necrosis.
